Background: The gingiva, a specialized part of the oral mucosa, is prone to frequent injury and exhibits a distinct wound healing process compared with other oral tissues. Caffeic acid phenethyl ester (CAPE), the primary bioactive compound in propolis, is known for its anti-inflammatory properties and its ability to enhance re-epithelialization. However, its role in modulating fibroblast activity and COL1A2 expression during gingival wound healing remains unexplored. Thus, the objective of this study was to investigate the COL1A2 expression in fibroblast underlying the effects of CAPE on gingival wound healing.
Materials and Methods: Forty-five male Balb/c mice were divided into three groups: control (saline), CAPE pre-treatment, and CAPE treatment. The CAPE was given through intraperitoneal (i.p) injection at a dose of 4 mg/kg body weight. Gingival lesions were created, and wound healing was assessed using clinical observations and histological analyses, including hematoxylin-eosin, toluidine blue, and immunohistochemistry COL1A2 staining, on days 0, 3, and 5.
Results: Histological analyses and clinical results revealed that CAPE administration significantly enhanced gingival wound closure in the pre-treatment and treatment groups compared with controls. The COL1A2 expression in fibroblast was notably increased in the CAPE-treated groups, suggesting accelerated fibroblast activity and tissue repair.
Conclusion: CAPE promotes gingival wound healing by enhancing fibroblast activity and COL1A2 expression, indicating its potential as a therapeutic agent for oral tissue repair.
